Optimized DNA sequence  encoding mature chain of Mouse alpha 1 antitrypsin -A1AT (GLU25-LYS413) including a C-terminal His tag was expressed in HEK293 cells.

Molecular weight:

Recombinant Mouse alpha 1 antitrypsin (SERPIN A1a) is a protein consisting of 396 amino acid residue subunits, due to glycosylation migrates as an approximately 55kDa band protein on reduced SDS-PAGE.

>95%, as determined by SDS-PAGE and HPLC
Biological activity:
The activity was tested by the ability to inhibit trypsin cleavage of a fluorogenic peptide substrate Mca-RPKPVE- Nval-WRKDnp-NH2, with a calculated ED50 < 4.5nM
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

Recombinant Mouse alpha 1 antitrypsin is lyophilized  from 0.2 μm filtered PBS solution,  pH7.2 ,  5% Trehalose.

A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

Recombinant Serpin A1 can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

alpha 1 antitrypsin

AAT is a 52-kDa serine protease inhibitor synthesized primarily in the liver and secreted into the circulation with a half-life of 45 days. AAT is the most abundant circulating serine protease inhibitor. The main substrate for AAT is neutrophil elastase, a broad-specificity serine protease secreted by neutrophils during inflammation. AAT is synthesized and secreted by the liver and functions primarily in the lungs, where it protects pulmonary tissue from degradation by neutrophil elastase. A deficiency of AAT results in promiscuous activity of neutrophil elastase, resulting in early onset emphysema and chronic obstructive pulmonary disease
