Optimized DNA sequence  encoding extracellular domain of human IGF-1R (CD221) including a C-terminal 6His tag was expressed in HEK293 cells.

 
Molecular weight:

Recombinant human IGF-1R (CD221) is a heterodimer protein, due to glycosylation migrates as an approximately 50kDa,120kDa,150kDa protein under reducing ocnditions on SDS-PAGE.


 
Purity:
>95%, as determined by SDS-PAGE and HPLC

 
Biological Activity:
The activity of recombinant IGF-1R was measured by its ability to bind Human IGF-1 in a functional ELISA assay.

 
Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant IGF-1 receptor is supplied as a 0.2 μm filtered PBS solution, pH7.2.

              
Storage:

The lyophilized protein is stable for at least 2 years from date of receipt at -20° C.

Upon reconstitution, the protein can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months, addition of a carrier protein to your working aliquots (0.1% BSA or HSA) is recommended for long term storage 

Avoid repeated freeze/thaw cycles.

 
 
Reconstitution:

A quick spin of the vial followed by gently mixing in distilled water to prepare a stock solution concentration not less than 0.1 mg/mL.

Avoid vigorous shaking or vortex

 

IGF-1R

Insulin-like growth factor 1 receptor (IGF-1R/CD221) is a membrane-associated tyrosine kinase receptor that plays an important role in cell growth, transformation, and protection of cells from a variety of apoptotic stimuli. IGF-1R(CD221) is a tetrameric receptor consisting of two -subunits and two transmembrane β- subunits that are linked by disulfide bonds.The - subunits are extracellular and bind IGF. Each transmembrane β-subunit contains an intracellular tyrosine kinase domain. Binding of IGF-I or IGF-II ligand to IGF-1R leads to phosphorylation of three key tyrosine residues in the kinase domain, leading to phosphorylation of downstream substrates.
