Endothelial protein C receptor

The endothelial protein C receptor (EPCR) is a predominant vascular cell receptor. EPCR binds both protein C (PC) zymogen and the serine protease activated protein C (APC) with similar affinity, thereby localizing PC and APC to the cell surface. Although EPCR is considered a nonsignaling receptor because of its short (Arg- Arg-Cys) intracellular tail, it is increasingly recognized as key receptor in the anticoagulant and cytoprotective PC pathways by facilitating (A)PC’s interactions. Binding of PC to EPCR facilitates activation of PC to APC by the thrombin–thrombomodulin complex.APC acts as anticoagulant, and it also conveys cytoprotective functions that require EPCR and protease activated receptor 1 (PAR1), and include antiinflammatory and antiapoptotic activities and endothelial barrier- stabilizing effects.
