Molecular weight:

Recombinant human TFPI-2 is a protein consisting of 198 amino acid residue subunits,due to glycosylation migrates as an approximately 30kDa band protein on reduced SDS-PAGE.


 
Purity:
>95%, as determined by SDS-PAGE and HPLC
 
Biological activity:
The activity was tested by the ability to inhibit trypsin cleavage of a fluorogenic peptide substrate Mca-RPKPVE- Nval-WRKDnp-NH2, with a calculated ED50 < 2.5nM
 
Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant Human Tissue factor pathway inhibitor 2 is lyophilized from 0.2 μm filtered PBS solution,  pH7.2 ,  5% Trehalose.

              
Reconstitution:
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

 
Storage:
Recombinant Tissue factor pathway inhibitor  can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

Tissue factor pathway inhibitor 2

TF-induced coagulation is regulated by a specific inhibitor termed tissue factor pathway inhibitor (TFPI). The mechanism of inhibition by TFPI involves its binding first to factor Xa (FXa) and then to the FVIIa TF complex. TFPI is a multivalent serine protease inhibitor with 3 independently folded Kunitz- type proteinase inhibitor domains and a highly basic, positively charged carboxyl terminus known to bind heparin. The first Kunitz domain (K1) binds TF/VIIa, and the second Kunitz domain binds factor Xa. It is the formation of this quaternary TF-VIIa-TFPI-Xa complex that dampens ongoing coagulatio
