Optimized DNA sequence  encoding extracellular domain of NEPH1 (LEU48 - LEU525 ) including a C-terminal His tag was expressed in HEK293 cells.

Molecular weight:

Recombinant mouse NEPH1 is a protein consisting of 485 amino acid residue subunits,due to glycosylation migrates as an approximately 70kDa band protein on reduced SDS-PAGE.

Biological activity:
The activity was tested by the ability of immobilized recombinant mouse NEPH1 to bind mouse MS-1 endothelial cells, after 90 min 70% of cells will adhere.
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

Recombinant mouse NEPH1 is lyophilized  from 0.2 μm filtered PBS solution,  pH7.2 ,  5% Trehalose.

A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

Recombinant NEPH1 can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

NEPH1, a molecule recently identified in mice by a retrovirus-mediated mutagenesis screen, contains five extracellular immunoglobulin-like domains and is structurally related to nephrin (7). NEPH1 is abundantly expressed in the kidney, and disruption of the NEPH1 gene in mice results in effacement of glomerular podocytes, heavy proteinuria, and early postnatal death. Nephrin and Neph1 are structurally similar type I transmembrane proteins of the immunoglobulin superfamily. They directly interact in the plane of the plasma membrane via their cytoplasmic domains (4, 12, 25). In addition, the Nephrin extracellular domain interacts with itself and with Neph1 via a trans- interaction across the mature podocyte intercellular junction; for this reason, it has been presumed that these proteins mediate cell-cell adhesive interactions at this site
