Optimized DNA sequence encoding Mouse Interleukin-17A mature chain was expressed in Escherichia Coli.

Molecular weight:

Native Mouse Interleukin17A, generated by the proteolytic removal of the signal peptide and propeptide, the molecule has a calculated molecular mass of approximately 16kDa.

Recombinant Mouse IL-17A is a disulfide-linked monodimeric protein consisting of 137 amino acid residue subunits, and migrates as an approximately 16kDa protein under reducing conditions in SDS-PAGE.

Biological Activity:
The activity was measured by the ability to bind Human IL17RA in a functional ELISA assay with a linear range of 10-500 ng/ml.

Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

Recombinant Mouse Interleukin-17 was lyophilized from a 0.2 μm filtered PBS solution pH 7.5.

A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

The lyophilized protein is stable for at least 2 years from date of receipt at -20° C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2° - 8° C for one month, or at -20° C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

Interleukin-17 (IL-17 or IL- 17A) is the founder member of the IL-17 cytokine family, which shares 50% homology with IL-17F. IL-17 cytokines are produced by T- helper (TH)17 cells, γδ T cells, and other immune cells. IL-17A and IL-17F form homodimers and heterodimers as functional ligands. IL-17 cytokines bind to 2 receptors, IL-17RA and IL-17RC. IL-17RC forms a receptor heterodimer with IL-17RA. The receptor heterodimer is preferred by IL-17 ligands , thus, either IL-17RA knockout (KO) or IL- 17RC KO completely abolishes IL-17 signaling . IL-17A and IL-17F are key proinflammatory cytokines involved in many inflammatory and autoimmune diseases.
