Optimized DNA sequence encoding Human MIP-3 (CCL23) mature chain was expressed in Escherichia Coli.

 
Molecular weight:

Human CCL23, generated by the proteolytic removal of the signal peptide
and propeptide and has a calculated molecular mass of approximately 11kDa.

Recombinant MIP-3(CCL-23) is a disulfide-linked monomeric protein consisting of 100 amino acid residue subunits and migrates as an approximately 11 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.

>98%, as determined by SDS-PAGE and HPLC

 
Biological Activity:
Determined by its ability to chemoattract human T cells using a concentration range of 2.0-40.0 ng/ml.

Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant CCL23 was lyophilized from a 0.2 μm filtered 20mM PB,100mM NaCl solution pH 7.5.

 
Reconstitution:
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

 
Storage:
The lyophilized protein is stable for at least 2 years from date of receipt at -20° C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2° - 8° C for one month, or at -20° C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

CCL23

CCL23 (MPIF-1, CK8, SCYA23), a member of the CC chemokine family, was originally isolated from a human aortic endothelial cell library and from the human monocytic cell line THP-1. It is most closely related to MIP-1 and interacts with its receptor CCR1, which is expressed on monocytes, dendritic cells ,lymphocytes, and endothelial cells. Functionally, CCL23 has chemotactic activity for monocytes, DC, lymphocytes, neutrophils, osteoclast precursor cells, and endothelial cells. In contrast, CCL23 reduces the proliferation of progenitor cells giving rise to granulocyte and monocyte lineages, whereas it enhances angiogenesis of endothelial cells
