Molecular weight
Recombinant CD34 is a monomer protein consisting of 270 amino acid residue subunits, due to glycosylation migrates as an approximately 50 kDa protein on SDS-PAGE.

>95%, as determined by SDS-PAGE and HPLC
Biological Activity
Measured by the ability to support the adhesion of the HUVEC cell line. Coated plates with immobilized recombinant Human CD34 ( 0.8 μg/mL) are able to adhere 40% of cells ( 4 x 104 cells/well) after one hour at 37 ℃
Endotoxin
Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).
Presentation
Recombinant CD34 is supplied as a 0.2 μm filtered PBS solution, pH7.2.
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers
Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20° C. Upon reconstitution, this cytokine can be stored in working aliquots at 2° - 8° C for one month, or at -20° C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.

Schematic outline of the experimental strategy.

  • Freshly isolated CD34+ umbilical cord blood cells were separated into four experimental groups: Group 1: non treated CD34+ cells analyzed immediately; Group 2: cells stimulated with cytokines only; Group 3: cells cultured with cytokines and undergoing a mock infection with polybrene only but no LV; Group 4: cells cultured with cytokines, then transduced with polybrene and LV.

Cord-derived human mast cell culture

  • We used the protocol of Dahl et al + stem cells were cultured for 2 weeks in StemSpan medium'>serum-free medium supplemented with 100 ng/ml human SCF, 50 ng/ml IL-6 and 1 ng/ml IL-3 (added during the first 14 days) and 100∶g/ml penicillin/streptomycin .
  • The medium was replenished with 10% FCS from week 8.
