Optimized DNA sequence  encoding  extracellular domain of Human Low density lipoprotein receptor (ALA22-ARG788) including a C-terminal His tag was expressed in HEK293 cells.

Recombinant LDL receptor was further labled by Biotin for detection by avidin conjugated secondary probes.

 
Molecular weight:

Recombinant LDL receptor is a  protein consisting of 773 amino acid residue subunits,  due to glycosylation migrates as an approximately 100kDa(mature form) and 140kDa(immature form) protein on SDS-PAGE.


 
Purity:
>95%, as determined by SDS-PAGE and HPLC
 
Biological Activity:
The activity was tested by the ability of immobilized recombinant Human PCSK9 to bind biotinylated recombinant human LDL Receptor in ELISA.  The ED50 was calculated to be 0.6ug/ml.
 
 
Endotoxin:
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/µg(1EU/µg).

 
Presentation:
Recombinant Human Low density lipoprotein receptor is lyophilized  from 0.2 μm filtered PBS solution,  pH7.2 ,  5% Trehalose.

              
Storage:
Recombinant LDL receptor can be stored in working aliquots at 2° - 8° C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

Low density lipoprotein receptor

The low density lipoprotein receptor (LDLR) binds with high affinity to the apolipoprotein (apo)B100 and apoE components of circulating LDL and VLDL, respectively, via a ligand- binding domain consisting of seven cysteine-rich repeats (R1R7). Following LDLR- mediated endocytosis, bound lipoproteins are released within the acidic environment of early endosomes and the LDLR recycles to the cell surface. Lipoprotein release and LDLR recycling are facilitated by a pH- dependent open (ligand- active) to closed (ligand- inactive) conformational switch involving an intramolecular interaction between the R4/R5 repeats of the ligand-binding domain and a β-propeller region in the epidermal growth factor (EGF) precursor homology domain of LDLR. LDLR- mediated uptake of LDL in the liver is the primary means of plasma LDL clearance and thus influences steady-state plasma LDL-cholesterol (LDL- C) levels and associated risk of coronary artery disease
